The familial nature of DMD was noted very early on by both Meryon and Gowers. In fact, Gowers recognized that the disorder was limited to males and transmitted by healthy females, a mode of inheritance now recognized to be that of an X-linked recessive trait. This chapter describes the mode of inheritance and mutation rate when considering the genetics of DMD.
