Numerous experimental studies indicate that HIV proteins have the capacity to injure neurons and thus may contribute to neuronal demise in the brains of HIV-infected individuals. Potentially neurotoxic viral proteins include the envelope proteins (gp120 and gp41), Vpr, Tat and Nef. In this section we will first give a brief general overview of the possible sources of neurotoxic HIV proteins in the brain, the experimental models used to study their neurotoxicity and current concepts of the contributions of these viral proteins to neuronal injury. In the second section we will discuss the experimental evidence supporting neurotoxic activities of the individual HIV proteins indicated above in more detail and summarize recent developments.
